Overview
KPV is derived from the C-terminal sequence of α-MSH, a peptide hormone involved in immune modulation and inflammatory signaling. Preclinical research has examined KPV for its effects on pro-inflammatory mediators including TNF-alpha, NF-κB, and MAP kinase activity. A notable feature studied in preclinical models is its apparent selective activity in inflamed tissue, attributed in part to uptake via PepT1 — a peptide transporter that is upregulated during intestinal inflammation.
Unlike full-length α-MSH, KPV does not appear to induce melanin production in preclinical settings. This profile has been examined in research seeking to isolate the effects of α-MSH-derived sequences from their pigmentation-related activity. Studies have examined KPV in models of intestinal inflammation, skin injury, and wound repair.

Intestinal Inflammation Research
Preclinical models of intestinal inflammation have been used to study KPV’s effects on inflammatory markers and mucosal tissue. Studies in murine colitis models have examined changes in myeloperoxidase (MPO) activity, inflammatory cell infiltration, and levels of TNF-alpha in colonic tissue following KPV administration. Research has also explored nanoparticle-based delivery systems functionalized with hyaluronic acid as a method for targeted colonic delivery, with preclinical data examining TNF-alpha suppression and mucosal tissue responses in these settings.
Skin and Wound Repair Research
Preclinical wound models have examined KPV for its effects on inflammatory markers, collagen metabolism, and IL-8 levels in injured tissue. Studies have also investigated its activity against Staphylococcus aureus and Candida albicans in experimental infection models. Research in these settings has examined how KPV modulates the inflammatory phase of wound repair and its potential effects on fibrotic tissue responses.
